Improved Biomolecule Retention

Magnify retains multiple biomolecule classes within the polymer gel, including protein, lipids, and nucleic acids. These biomolecules are all available for post-expansion labeling and imaging.

Broadly Applicable Across Tissues

Magnify is able to expand previously challenging FFPE tissues without the use of harsh protease, allowing post-expansion biomolecule profiling in a wide range of samples.

A reformulated gel allows 8 to 11-fold expansion depending on tissue type. The Magnify gel is highly robust and easy to handle even when fully expanded.

Exciting news! Magnify has been featured on the cover of the June 2023 issue of Nature Biotechnology!

 

Magnify is a new expansion microscopy protocol which uses a mechanically sturdy gel that retains nucleic acids, proteins, and lipids without the need for a separate anchoring step. We demonstrated up to 11-fold expansion in a wide variety of tissues, including FFPE human samples. Magnify is available online here. Congratulations, team!

Magnify was put to the test on a multiple tissue array of FFPE human samples, and was able to expand all tissue cores without cracks or distortion after homogenization in a hot surfactant solution.

Lipid molecules were retained in these Magnify-expanded HEK cells without requiring custom anchoring molecules, enabling the visualization of its membranes with post-expansion application of the lipophilic dye DiD.

11-fold expansion allows synaptic contacts to be studied with remarkable detail. The synaptic cleft, mitochondria, and individual vesicles can all be identified in this synapse onto a cortical neuron.
